Use Distinct Pathways to Repress Telomerase Expression

چکیده
Background: Inactivation of p53 and reactivation of telomerase expression are two of the most common events in human cancer. Results: Here we report the mechanisms used by p53, p63, and p73 to suppress telomerase expression. Conclusion: p53, p63, and p73 suppress telomerase expression through distinct pathways that involve E2F, E-box, c-Myc, and NF-YB. Significance: This study provides new insights into the cellular pathways used by p53, p63, and p73 to suppress hTERT expression.

Multiple Tumor Suppressor Pathways Negatively Regulate Telomerase
Telomerase expression is repressed in most somatic cells but is observed in stem cells and a high percentage of human cancers and has been hypothesized to contribute to tumorigenesis and maintenance of stem cell states. To explore telomerase regulation, we employed a general genetic screen to identify negative regulators of hTERT. We discovered three tumor suppressor/oncogene pathways involved ...
متن کاملCaenorhabditis elegans POT-1 and POT-2 Repress Telomere Maintenance Pathways
Telomeres are composed of simple tandem DNA repeats that protect the ends of linear chromosomes from replicative erosion or inappropriate DNA damage response mechanisms. The mammalian Protection Of Telomeres (POT1) protein interacts with single-stranded telomeric DNA and can exert positive and negative effects on telomere length. Of four distinct POT1 homologs in the roundworm Caenorhabditis el...
متن کاملAttenuation of telomerase activity by a hammerhead ribozyme targeting the template region of telomerase RNA in endometrial carcinoma cells.
Telomerase activity is found in almost all carcinoma cells but not in most somatic cells, suggesting that telomerase is an excellent target for cancer therapy. We designed hammerhead ribozymes against human telomerase RNA and studied their possible use as a tool for cancer therapy. Three ribozymes targeting the 3' end of the GUC sequence at 33-35 (the template region), 168-170, and 313-315 from...
متن کاملNanocurcumin-Mediated Down-Regulation of Telomerase Via Stimulating TGFβ1 Signaling Pathway in Hepatocellular Carcinoma Cells
Background: Curcumin, extracted from turmeric, represents enormous potential to serve as an anticancer agent. Telomerase is viewed as a prominent molecular target of curcumin, and transforming growth factor-β1 (TGFβ1) has proven to be a major inhibitory signaling pathway for telomerase activity. In the current study, we aimed to explore suppressive effects of nanocurcumin on telomeras...
متن کاملThe Wilms' tumor 1 tumor suppressor gene represses transcription of the human telomerase reverse transcriptase gene.
Regulation of the human telomerase reverse transcriptase (hTERT) gene is the primary determinant for telomerase enzyme activity, which is found in tumor cells but is largely absent from normal somatic cells. Recent studies have shown that Myc protein can transcriptionally activate the hTERT gene. However, little is known about the repression mechanism of the hTERT gene and telomerase enzyme. He...
